Öğe 2,6-Bis((E)-((5-benzoyl-2-thioxo-4-phenylpyrimidin-1(2H)-yl)imino)methyl)-4-(methyl)phenol and its Metal(II) Complexes: Synthesis, Spectroscopy, Biological Activity, and Photoluminescence Features(Wiley-V C H Verlag Gmbh, 2013) Gulcan, Mehmet; Zengin, Huseyin; Celebi, Metin; Sonmez, Mehmet; Berber, IsmetThe preparation, characterization, magnetic measurements, antimicrobial activity, and photoluminescence properties of a number of metal complexes with a Schiff base ligand derived from 2, 6-diformyl-4-methylphenol and 1-amino-5-benzoyl-4-phenyl-1H-pyrimidine-2-thione are reported herein. UV/Vis spectra and magnetic measurements infer square planar stereochemistry for Pd-II and Pt-II complexes, and octahedral arrangement for the other metal complexes. The compounds results in intense emission (where (max) = 402 nm) upon irradiation by UV light. The photoluminescence quantum yields and long excited-state lifetimes of the ligand and its complexes were calculated as 42% and 3.94 ns, respectively. The photoluminescence intensities and quantum yields of metal complexes changed upon complexation with various metal ions. The ligand and its complexes are of interest as organic emitting materials for electroluminescent devices. The complexes were additionally evaluated for in vitro the antimicrobial activity against bacterial and fungal strains. While Cu-II, Co-II, and Pt-II complexes have good antifungal efficiency against yeast (MICs, 80 gmL(-1)), only the complex [Cu(L)(AcO)]4H(2)O had effective and selective antimicrobial activity against all tested microorganisms with MIC values in the range of 40-80 gmL(-1), except for E. coli ATCC 4230. This study suggests that the complex [Cu(L)(AcO)]4H(2)O may be a new potential antimicrobial substance towards bacteria and fungi.Öğe A new pyrimidine-derived ligand, N-pyrimidine oxalamic acid, and its Cu(II), Co(II), Mn(II), Ni(II), Zn(II), Cd(II), and Pd(II) complexes: synthesis, characterization, electrochemical properties, and biological activity(Taylor & Francis Ltd, 2010) Sonmez, Mehmet; Celebi, Metin; Levent, Abdulkadir; Berber, Ismet; Senturk, ZuehreA new heterocyclic compound N-(5-benzoyl-2-oxo-4-phenyl-2H-pyrimidin-1-yl)-oxalamic acid has been synthesized from N-amino pyrimidine-2-one and oxalylchloride. Bis-chelate complexes of the ligand were prepared from acetate/chloride salts of Cu(II), Co(II), Mn(II), Ni(II), Zn(II), Cd(II), and Pd(II) in methanol. The structures of the ligand and its metal complexes were characterized by microanalyses, IR, AAS, NMR, API-ES, UV-Vis spectroscopy, magnetic susceptibility, and thermogravimetric analyses. An octahedral geometry has been suggested for all the complexes, except for Pd(II) complex, in which the metal center is square planar. Each ligand binds using C(2)=O, HN, and carboxylate. The cyclic voltammograms of the ligand and the complexes are also discussed. The new synthesized compounds were evaluated for antimicrobial activities against Gram-positive, Gram-negative bacteria and fungi using the microdilution procedure. The Cu(II) complex displayed selective and effective antibacterial activity against one Gram-positive spore-forming bacterium (Bacillus cereus ATCC 7064), two Gram-positive bacteria (Staphylococcus aureus ATCC 6538 and S. aureus ATCC 25923) at 40-80 mu g mL-1, but poor activity against Candida species. The Cu(II) complex might be a new antibacterial agent against Gram-positive bacteria.Öğe Antimicrobial and antioxidant activities of medicinal plant Glycyrrhiza glabra var. glandulifera from different habitats(Taylor & Francis Ltd, 2016) Karahan, Faruk; Avsar, Cumhur; Ozyigit, Ibrahim Ilker; Berber, IsmetIn this study, the antimicrobial and antioxidant activities of root methanolic extracts of Glycyrrhiza glabra var. glandulifera (Waldst. & Kit.) Boiss. (Fabaceae) were investigated. Plant samples were collected from different habitats in the East Mediterranean part of Turkey. The plant extracts were evaluated for antimicrobial activities against nine bacterial and two yeast strains using disc-diffusion and minimum inhibitory concentration methods. The antioxidant activity was determined by using the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. The antimicrobial assays indicated that the plant root extracts were more effective against Gram-positive bacteria than against Gram-negative ones. In addition, the extracts had higher antimicrobial effect against Candida species than against bacteria. The extracts showed good antioxidant activity, with a median inhibitory concentration (IC50) in the range of 588 +/- 0.86 mu g/mL to 2190 +/- 1.73 mu g/mL. Results indicated that different environmental conditions in each habitat might affect the contents of chemical compounds and biological activity in the natural licorice populations of. This study also supported the traditional use of licorice and as well as suggested that it may also be its beneficial role in the treatment of other infections. The obtained results indicated that different environmental conditions in each habitat might affect the contents of chemical compounds and the biological activity in the natural licorice populations.Öğe Characterization of Environmental Bacillus Isolates by Protein Fingerprinting and Random Amplified Polymorphic DNA Profiles(Springer India, 2017) Berber, Ismet; Avsar, Cumhur; Yegin, Zeynep; Civek, SeyhanThe aim of the present study was to investigate antibiotic resistance, plasmid, random amplified polymorphic DNA (RAPD) and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) profiles of the Bacillus spp. isolated from soil of Sinop environs, Turkey. A total of fifty-seven isolates were identified as Bacillus spp. based on morphological and physiological properties. All isolates were found resistant to bacitracin. Susceptibility patterns of the isolates to other tested antibiotics varied from 100 to 31.5 %. The plasmid analysis results indicated that isolates harbored 2 to 7 plasmids. The numerical analysis of whole cell and extracellular protein profiles of the isolates displayed similarity levels between 78–100 and 67–100 % respectively reflecting the less discriminative power of SDS-PAGE at subspecies level. However, RAPD analysis of genomic DNA using four different universal primers revealed 100 % polymorphism indicating the strength of RAPD in the differentiation of Bacillus isolates. © 2016, The National Academy of Sciences, India.Öğe Investigating Some Microbial Pollution Parameters of Seawater and Mussels (Mytilus galloprovincialis, Lamarck 1819) of Sinop Black Sea Coastal Zone, Turkey(Univ Kebangsaan Malaysia, 2014) Berber, Ismet; Avsar, CumhurThe presence of coliforms bacteria is one of the most prevalent problems in terms of public health in marine ecosystems over the world. In this study were investigated the physico-chemical properties of seawater and the numbers of total aerobic, total coliform, fecal coliform, E. coli O157: H7 and fecal streptococci in seawater and mussel samples collected from Sinop environs between May and October 2011. The microbiological analysis of seawater samples showed that the difference between total coliform, fecal coliform and fecal streptococci numbers (p< 0.05) was significant for each station. However, the difference among total aerobic bacteria numbers for each stations (p> 0.05) were not found significant. The difference between whole counting results for mussel samples taken from different sampling sites was not significant (p> 0.05), too. Furthermore, the results of the screening assay for the presence of E. coli O157: H7 showed that the strain was not detected in neither seawater nor mussel samples. In conclusion, it was determined that fecal coliform and fecal streptococci counts in the seawater and mussel samples were higher than legal (Turkish Bathing Water and Quality of Fishery Products Regulation) limit values for some stations in Sinop coastal areas.Öğe Isolation and characterization of Vibrio spp. from anchovy and garfish in the Sinop province(Refik Saydam National Public Health Agency (RSNPHA), 2016) Avşar, Cumhur; Berber, Ismet; Yildirim, Ahmet KenanObjective: In this study it was aimed to characterize the Vibrio species isolated from anchovy and garfish samples obtained from fishermen and to determine the antibiotic resistances, plasmide DNA and SDS-PAGE (sodium dodecyl sulphate polyacrylamide gel electrophoresis) whole cell protein profiles of these isolates. Method: The fish samples were taken as fresh, kept in ice during transport to the laboratory and immediately taken for analysis. The fish samples weighed as 25 grams were added into 225 mL of sterile alkaline peptone water supplemented with 3% NaCl broth for enrichment purpose and incubated at 37 OC for 18 to 24 hours. After incubation, yellow, green or blue-green colonies growing on Thiosulphate Citrate Bile Salt Sucrose (TCBS) agar were subcultured on Triptone Soya Agar (TSA) and incubated overnight for further identification of the isolates. The isolates were identified according to morphological, physiological and biochemical tests. Moreover, the resistances against seven different antibiotics of isolates were determined by disk-diffusion method. In addition, the SDS-PAGE cell protein and plazmid DNA profiles of isolates were determined. Results: Totallly 44 fish samples were tested for determining Vibrio species and were analyzed according to their morphological, biochemical properties. At the end of the analysis; 12 Vibrio species were determined. The isolates were resistant against to ceftazidime 66.6%, gentamisine 25%, tetracycline 16.6%, ceftriaxone 91.6%, amikacine 16.6%, ofloxacine 25% and penicillin G 58.3%. In addition, The MARI (Multiple Antibiotic Resistance Index) values of tested strains were found at the rate of 0.41. The numerical analysis of SDS-PAGE whole-cell protein profiles of Vibrio spp. isolates revealed two major clusters at similarity levels of 80% and above. The 1-4 plasmide (range 1590 - 27000 bp) in all of the isolates were determined. Conclusion: As a result, it is possible to say that microbial infections can be prevented by adequate freezing and cooking, proper storage and processing after harvesting and avoidance of cross-contamination during fish handling. In addition, the coastal contamination due to bacteria leads to decline in the quality of seawater resources and so leads to risks to human health and economic losses. Furthermore, our study indicated that the presence of antibiotic resistant Vibrio spp. strains isolated from fishes might be potential risk for health.Öğe Lack of association between MAOA-uVNTR variants and excessive daytime sleepiness(Springer-Verlag Italia Srl, 2017) Ozen, Filiz; Yegin, Zeynep; Yavlal, Figen; Saglam, Zuhal Aydan; Koc, Haydar; Berber, IsmetSleep disorders are highly prevalent in the population and have dramatic health, social, and economic impacts. However, their treatments may remain symptomatic due to ignorance of molecular factors which may provide fundamental insights into the neurological bases of sleep. Excessive daytime sleepiness (EDS) is a common complaint encountered in neurological practice with significant effects both on individuals and on society. We aimed to investigate the role of monoamine oxidase A (MAOA) as a candidate gene in EDS. Epworth sleepiness scale (ESS) was applied to 221 subjects who were also genotyped for MAOA upstream variable number of tandem repeats (MAOA-uVNTR). Patient group displayed higher ESS values (mean 12.67) when compared with the control group (mean 6.38). However, MAOA-uVNTR genotypes did not show a significant association with ESS scores neither on women nor on men. Finally, these data suggest further replications in different populations. Moreover, the investigation of some other genes together with MAOA and/or some possible regulatory molecular mechanisms may offer a more comprehensive approach in the role of genetic factors contributing to EDS.Öğe Levels of Selected Trace Elements, Phytohormones, and Sugars in Pseudomonas-Infected Lycopersicum esculantum Mill Plants(Humana Press Inc, 2010) Berber, Ismet; Ekin, Suat; Battal, Peyami; Onlu, Harun; Erez, M. EmreThe present study investigated the levels of trace elements (Zn, Cu, Fe, Pb, and Cd), major elements (Ca and K), phytohormones (trans-Zeatin [t-Z] and gibberellic acid [GA]), and sugars (sucrose and glucose) following inoculation with Pseudomonas syringae pv. tomato strain. The results of the trace elements analysis showed that Fe (in the first, fourth, eighth, and tenth study days), Cd (in the fourth, eighth, and tenth study days), Cu (in the fourth and eighth study days), and Zn (in the eighth and tenth study days) increased in bacterium-infected tomato plants, compared to healthy plants. The levels of Pb, Ca, and K did not meaningfully determine a change after inoculation with pathogen. In this vein, the increase accumulation rates of Cu, Zn, and Fe in the injured plants can be an important indicator for the plant defense processes towards pathogen attack. Furthermore, in the first, fourth, eighth, and tenth study days, the glucose and sucrose contents crucially decreased in bacterium-infected plants compared to the control groups. The lowest level of sucrose in bacterium-infected plants was observed on the first day. The findings displayed that, when endogenous t-Z levels did not change after inoculation with virulent bacterium strain, there was a reduction in the first, fourth, eighth, and tenth days in the level of GA when compared with the control group levels. Therefore, there may be a link between lower GA level and enhancement in the expression of defense-related genes. The results of this study showed that there are complex relationships among levels of sugar, trace element, and endogenous phytohormone in the regulation of defense mechanisms against bacterial pathogen attacks.Öğe Method validation and simultaneous determination of retinol, retinyl palmitate, β-Carotene, α-Tocopherol and vitamin C in Rat serum treated with 7,12 dimethylbenz[a]anthracene and plantago major L. High-performance liquid chromatography analysis was performed utilizing an Inertsil ODS3 reversed phase column with methanol-tetrahydrofuran-water as mobile phase under gradient conditions, at 1.5 mL min-1 flow rate and 25 °C. Diode-array detection was at 325, 450, 290 and 270 nm (retinol and retinyl palmitate), β-carotene, α-tocopherol and vitamin C, respectively and runnig time 18 min. The highperformance liquid chromatography assay and extraction procedure proposed are simple, rapid, sensitive and accurate. The method was then applied for the determination of retinol, retinyl palmitate, β-carotene, α-tocopherol and vitamin C in rat serum. Results of this study demonstrated that; at 60th day DMBA-treated group, there was a significant decrease in vitamin levels compared to the levels of control group. A significant increase was observed in vitamin levels of 7,12 dimethylbenz[a]anthracene+Plantago Major L.-treated group compared to the DMBA-treated group. The acetylation of compound 1a gave 3-acetyl thioxopyrimidin 2. Also, thiazolopyrimidine (3) and (4) derivatives were obtained by a simple one-pot condensation reaction of starting compounds 1a, and 1b with 2-bromopropionic acid. The prepared compounds were screened for their antimicrobial activities against Gram-positive, Gram-negative bacteria and fungi using micro dilution procedure. The results of the study showed that the compound 4 was effective and selective for antimicrobial activity against the tested bacteria and fungi. Methods: Rats were divided into three equal groups of 6 animals each: Group 1 controls, group 2 treated with DMBA (100 mg/kg, single dose) and group 3 receiving the DMBA and the aqueous extract at 100 mg/kg/d for 60 days. Results: Significant decrease in catalase (P<0.05), carbonic anhydrase (p <= 0.01), reduced glutathione (GSH) (P<0.01) and total protein (P<0.01) values was observed in the DMBA group compared with the healthy controls and DMBA + Plantago major groups. Octahedral geometry was suggested for all the complexes, in which the metal center coordinates to ONO donors of the ligand. Each ligand binds the metal using C=O, HN, and carboxylate. The cyclic voltammograms of the ligand and the complexes were also discussed. The compounds were evaluated for their antimicrobial activities against Gram-positive and Gram-negative bacteria, and fungi using microdilution procedure. The antimicrobial studies showed that Cu(II), Fe(III), and Ru(III) complexes exhibited good antibacterial activity against Gram-positive bacteria with minimum inhibitory concentrations between 20 and 80 mu g mL-1. However, the ligand and the complexes possess weak efficacy against Gram-negative bacterium and Candida strains. The complexes were prepared by reacting ligand and the metal chlorides of Cu (II), Ni(II), Co(II) and Fe(II) in methanol to get a series of mononuclear complexes. The complexes were characterized by elemental analyses, conductivity measurements, magnetic susceptibility data, IR, UV-Vis, NMR and API-ES mass spectral data. The mononuclear structure of the complexes was confirmed on the basis of elemental analyses, magnetic susceptibility and API-ES mass spectral data. The ligand and all the metal complexes were evaluated for their antimicrobial activity against four Gram-positive bacteria (Staphylococcus aureus ATCC 6538, S. aureus ATCC 25923, Bacillus cereus ATCC 7064 and Micrococcus luteus ATCC 9345), one Gram-negative bacterium (Escherichia coli ATCC 4230), and three yeast (Candida albicans ATCC 14053, Candida krusei ATCC 6258 and Candida parapsilosis ATCC 22019) strains. All rights reserved.Öğe The changes in the serum, liver, kidney protein and genomic DNA profiles in rats treated with 7,12-Dimethylbenz(A) anthracene and Plantago major L.(2014) Berber, Ismet; Ekin, Suat; Levent, Abdulkadir; Oto, GokhanIn the study, the changes in the serum, liver, kidney protein and genomic DNA profiles in Wistar albino rats administrated to 7,12-dimethylbenz(a)anthracene (DMBA) and Plantagomajor L. were investigated. SDS-PAGE profiling of the serum proteins showed that the levels of two proteins molecular weighing 140.8 kDa and 46.8 kDa were significantly lower on the 0th day of treatment than on the 60th day of application in the groups. Furthermore, two proteins (34.5 and 22.3 kDa) were solely present in DMBA-treated liver cell lysates. On the contrary, the kidney protein profiles did not show significant banding variations in groups. Genomic DNA analysis also confirmed that DNA extracted from liver and kidney cells did not fragment in any of the groups after the 60th study day. The proteins 34.5 and 22.3 kDa in liver cell lysates at the 60th day in DMBA-treated group could be extra-prognostic indicators for severe DMBA toxicity.Öğe The Determination of Antimicrobial Activities of Some Plants Growing in Van Region(Van Yüzüncü Yil Üniversitesi, 2009) Berber, Ismet; Özgökçe, Fevzi; Seker, AyseIn the present study, the antimicrobial activities of 10 different plant species growing in Van region were determined against 5 Gram-positive (Staphylococcus aureus ATCC 25923, S. epidermidis, Micrococcus iuteus ATCC 9345, Bacillus cereus ATCC 7064 and B. thuringiensis var. israelensis 2 Gram-negative (Escherichia coli ATCC 4230 and Enterococcus faecalis ATCC 29212) and 3 yeast (Candida krusei ATCC 6258, C. parapffosis ATCC 22019 and C. albicans ATCC 14053) $trains by using disc diffusion method. White the extracts of Saivia kronenburgii, Antriseus nenorosa and Cephaloria setosa were the highest antimicrobial activity against all tested strains, it was found that the extracts of Thymus transcaucasjcusj Morus albay Pinus nigra subsp. pallasiana and Hypericum perforatum had no antimicrobial activity. However, the extracts of Syrjngae vulgaris and Salix alba exhibited Iow antimicrobial activity against tested microorganisms. Also the extract of Alyssum meniocoides was most effective against yeast strains (1216 mm). Consequently, the research indicated that Tris-HCI extracts of four plants extract that having good antibacterial and antifungal activity can be used as natural antimicrobial agents.
