Öğe Evaluation of CDH1 Promoter Methylation and HPV Infection Status in the Development of Parotid Pleomorphic Adenoma(Kamla-Raj Enterprises, 2020) Ozen, F.; Yegin, Z.; Acar, G. O.; Avsar, C.; Zenginkinet, T.Both benign and malignant salivary gland tumors are clinically heterogeneous and they display different histology. Researchers aimed to elucidate the potential roles of both CDH1 methylation and high-risk oncogenic human papilloma virus (HPV) types HPV16 and HPV18 in parotid pleomorphic adenoma. Genomic DNA samples extracted from fonnalin-fixed paraffin-embedded (FFPE) samples of parotid pleomorphic adenoma and control tissues were subjected to methylation specific PCR (MSP) for hypermethylation analyses. HPV infection presence was evaluated using both GP5+/GP6+ consensus primers and type-specific primers for IIPVI6 and HPVI8. Both CDH1 promoter hypermethylation and HPV16 infection partially seemed to be responsible in the occurrence of the tumors. According to the best of the researchers' knowledge, this is the first study investigating the possible roles of both CDH1 methylation and HPV involvement in parotid pleomorphic adenoma. Multicentered studies with larger sample cohorts could be highly beneficial to strengthen this study's results and thus draw a more precise conclusionÖğe Roles of p53 Codon 72 and miR-502-binding Site in the 3′-UTR of SET8 SNPs in Urinary Bladder Cancer Predisposition in Turkish Population(Kamla-Raj Enterprises, 2019) Yegin, Z.; Ozen, F.; Koc, H.; Ulus, I; Yildirim, A.; Buyukalpelli, R.A case-control study was conducted to elucidate the possible roles of two SNPs (TP53 codon 72 and SET8 miR-502) that may affect each other in the same signaling pathway in bladder cancer susceptibility. Genotype distributions of 180 bladder cancer patients in comparison with 203 cancer-free controls were tested using PCR-RFLP method. For TP53, the researchers observed some protective effect of CC (Pro/Pro) genotype when compared with the heterozygous form (CG). For SET8 miR502, the researchers found no significant association in terms of variant alleles. These data suggest the association of TP53 codon 72 SNP, but not SET8 miR-502 with bladder cancer susceptibility in Turkish population. Further studies with larger sample sizes and different ethnicities are desirable to validate the researchers' findings.Öğe Thymoquinone Down-regulates VEGFA and Up-regulates FLT1 Transcriptional Levels in Human Breast Cancer Cells(Kamla-Raj Enterprises, 2020) Yegin, Z.; Duran, T.; Yildirim, I. H.Angiogenesis is important for cancer progression index and angiogenesis factors related to tumorogenesis deserve to be investigated in detail. The use of minimally toxic phytochemical compounds as the new generation anticancer agents is an appreciated approach to manage angiogenesis factors. The purpose of this study was to investigate the potential effects of thymoquinone (TQ), the major constituent of the black seed, on the expression levels of VEGFA and its receptor FLT1 in human estrogen receptor-positive breast adenocarcinoma (MCF-7) cells. The researchers provide evidence that TQ down-regulated VEGFA and up-regulated FLT1 transcriptional levels in human breast cancer cells compared to HEK293 cells. To the best of the researchers' knowledge, this is the first study determining the effect of TQ in VEGFA and its receptor FLT1 in MCF-cells and more comprehensive investigations are highly recommended.
